The European Commission works like a government and an administration. Politicians change every 5 years after the elections, or even more. The administration works like another administration: it does what the politicians decide what they should do.
How do you get in the political Commission? You need to convince one of the EU's countries' governments you are the right candidate. They will propose you (or another person from that country). The Parliament will test you. Once you've passed the test, you can start working there.
What kind of work will you be doing? Proposing laws, updating laws, spending the budget... mostly what other governments do. Every Commissioner has a specific area in which they work: trade, energy, labor...
